چکیده

We describe an apparatus to measure the diffusion of dilute fluorophores in molecularly thin liquid films within a surface forces apparatus ~SFA!. The design is a significant modification of the traditional SFA in that it allows one to combine nanorheology with the single-molecule sensitive technique of fluorescence correlation spectroscopy. The primary enabling idea was to place a miniaturized SFA onto the stage of an optical microscope equipped with a long working distance objective and illuminated by a femtosecond laser. A secondary enabling idea was that the silver coating on the backside of mica, normally used in the traditional SFA design for interferometric measurements of the film thickness, was replaced by multilayer dielectric coatings that allowed simultaneous interferometry and fluorescence measurements in different regions of the optical spectrum. To illustrate the utility of this instrument, we contrast the translational diffusion of rhodamine dye molecules ~in the solvent, 1,2-propane diol!, in the unconfined bulk state and confined between mica sheets to the thickness 2.5 nm. The diffusion coefficient is found to decrease by 2 orders of magnitude under confinement. © 2003 American Institute of Physics. @DOI: 10.1063/1.1570947#

Hyperbranched Polymer Integrated Membrane for the Removal of Arsenic(III) in Water

This work demonstrates the synthesis, characterization and application of a hyperbranched polyethyleneimine/polysulfone (HPEI/PSf) thin fi lm composite (TFC) membrane. The membrane was accessed via an interfacial polymerization of trimesoyl chloride and HPEI. The membrane samples were characterized by Fourier Transform Infrared-Attenuated Total Refl ectance (FTIR-ATR) s...

Integrated fluorescence correlation spectroscopy device for point-of-care clinical applications

We describe an optical system which reduces the cost and complexity of fluorescence correlation spectroscopy (FCS), intended to increase the suitability of the technique for clinical use. Integration of the focusing optics and sample chamber into a plastic component produces a design which is simple to align and operate. We validate the system by measurements on fluorescent dye, and compare the...

Decoupling geometrical and chemical cues directing epidermal stem cell fate on polymer brush-based cell micro-patterns.

The intricacy of the different parameters involved in cell adhesion to biomaterials and fate decision (e.g. proliferation, differentiation, apoptosis) makes the decoupling of the respective effects of surface properties, extra-cellular matrix protein adsorption and ultimately cell behaviour difficult. This work presents a micro-patterned polymer brush platform to control the adsorption of extra...

Hydrodynamic size-dependent cellular uptake of aqueous QDs probed by fluorescence correlation spectroscopy.

Aqueous quantum dots (QDs) directly synthesized with various thiol ligands have been investigated as imaging probes in living cells. However, the effect of the surface chemistry of these ligands on QDs' cellular uptakes and their intracellular fate remains poorly understood. In this work, four CdTe QDs were directly synthesized under aqueous conditions using four different thiols as stabilizers...
